Default Possible Transmission Problem

Hello, all, I need some information on my 2008 Silverado w/128000 mi. Recently here in charlotte we've had some cold weather, after cranking my truck I go to put the it in Drive and give it a little gas IT FEELS LIKE SOMETHING IS HOLDING THE TRUCK BACK, so i give it more gas and when it finally moves it's a LOAD CLUNKING SOUND. It don't happen evertime but enough for concern. Im in the process of getting the Slip Yolk changed but wanted to know if it could be the TRANSMISSION?
