Default Back Lash question 03 tahoe

Hi All,
03 Tahoe, When I let off the gas and go around a corner and apply the gas again, there is a slight clunk in the rear end. And when the rear tires go over a speed bump I can feel the looseness in the rear diff. These are the only times this happens. Otherwise it drives fine. In neutral and the break set, the drive line has play in it. In park it does not have any play at all. Is there supposed to be any play at all in the yoke at the diff? In my limited experience with a 72 Buick. The Back lash was not set correctly after I replaced the oil seal. I tightened up the yoke just a little and that fixed it. Any ideas?
