Default No start Stumped. need help

Iv got a 2001 Express 2500 with a 350. I had a no start condition but could here the fuel pump and had spark, when I would hit it with starting fluid nothing would happen which would mean no fuel issue. but I changed the cap and rotor because I know they are a common problem spot. Then tried starting and still no start so I git it with starting fluid once again and it fired up sputtered and died, so I fired it up again keeping my foot on the gas spittle then slowly letting off when it started then let it run for ten mins. I shut it off and started it a couple more times then left. The next day it wouldn't start again. Fuel pressure spec 60-66 psi with the fuel pressure gauge on actual fuel pressure 55 psi when key on engine off, left hooked up over night and this morning it had 27 psi. Hit key on and engine off right back to 55 psi.
Fuel pressure regulator? Fuel filter? I'm kinda stumped.

I put a new fuel filter on and the van started right up. Fuel pressure jumped around 55 psi. Next morning wouldn't start. Any ideas?
